Postural Orthostatic Tachycardia Syndrome (POTS) is a debilitating condition that affects between 0.2 and 1% of the US population and may be even more prevalent since the COVID-19 pandemic. The conditionimpairs individuals' ability to participate in routine activities such as working, attending school, exercising and activities of daily life. POTS primarily affects women of child-bearing age, with most studies reporting 80-90% female predominance. While many individuals with POTS report symptoms beginning in adolescence, up to half of all individuals with POTS develop it in adulthood. The impact of POTS across the lifespan has until now not been well studied, and given the female preponderance of the condition there is an unmet need to study how the condition is impacted by events such as menarche, pregnancy and menopause. There is a compelling need to stimulate research focused on improving diagnosis and treatment of POTS. This may include development of biomarkers and diagnostic tools as well as translational studies and mechanistic clinical trials to guide the development of interventions aimed at preventing and curing POTS. This NOSI signals interest in this important area with the goal of stimulating research applications to address these critical needs.

This Notice of Funding Opportunity (NOFO) supports the optimization of promising genome editing-based therapeutic leads for Alzheimer's Disease-Related Dementias (ADRD), towards IND-enabling studies. Specifically, it supports the characterization and optimization of therapeutic lead(s) that show promise as potential genome editing therapeutics, as evidenced by convincing proof-of-concept studies in appropriate models. At the end of the funding period, successful projects will have optimized a genome editing therapeutic candidate with demonstrated bioactivity, manufacturability, biodistribution, in vivo efficacy, and/or target engagement (measurement of proximal downstream effects) and optimal dosing, combined with other properties consistent with the desired clinical application. This NOFO is not specific for any one or group within the ADRD spectrum of disorders. Disorders include; Frontotemporal dementia (FTD), Lewy body dementias (LBD) (including Dementia with Lewy bodies (DLB), Parkinson Disease Dementia (PDD)), Vascular contributions to Cognitive Impairment and Dementia (VCID), and Multiples Etiology Dementias (MED).

The NINDS Materials to Enhance Training in Experimental Rigor (METER) UE5 will support curriculum development in the form of innovative educational materials that will be incorporated into a new cutting-edge online resource that aims to promote awareness, understanding, and practice of fundamental principles of rigorous biomedical research for researchers and other scientists in various career stages and learning environments.This UE5 FOA runs in parallel with a companion UC2 FOA that solicits applications for the NINDS center for Creating an Educational Nexus for Training in Experimental Rigor (CENTER), described in detail in RFA-NS-21-009.

The purpose of the MOSAIC Postdoctoral Career Transition Award to Promote Diversity (K99/R00) program is to support a cohort of early career, independent investigators from diverse backgrounds conducting research in NIH mission areas. The long-term goal of this program is to enhance diversity in the basic biomedical sciences research workforce. The MOSAIC K99/R00 program is designed to facilitate a timely transition of outstanding postdoctoral researchers from diverse backgrounds (e.g., see NIHs Interest in Diversity) from their mentored, postdoctoral research positions to independent, tenure-track or equivalent faculty positions at research-intensive institutions. The MOSAIC K99/R00 program will provide independent NIH research support during this transition to help awardees launch competitive, independent research careers. Additionally, MOSAIC K99/R00 scholars will be part of organized scientific cohorts that will be expected to participate in mentoring, networking, and professional development activities coordinated by MOSAIC Institutionally-Focused Research Education Award to Promote Diversity (UE5) grantees.
