Puerarin Attenuated Early Diabetic Kidney Injury through Down-Regulation of Matrix Metalloproteinase 9 in Streptozotocin-Induced Diabetic Rats

Abstract: Radix puerariae, a traditional Chinese herbal medication, has been used successfully to treat patients with early stage of diabetic nephropathy. However, the underlined mechanism of this renal protective effect has not been determined. In the current study, we investigated the effects and the mechanism of puerarin in Streptozotocin (STZ)-induced diabetic rats. We treated STZ-rats with either puerarin or losartan, an angiotensin II receptor blocker, as compared to those treated with vehicle. We found that both … Show more

[29][30][31] The previous study found that Puerarin may induce therapeutic angiogenesis in myocardium by inducing VEGF and eNOS expression in myocardium of rat with myocardial infarction.…”

“…12 It has been shown effective in the treatment of kidney diseases, such as nephrotoxicity, acute kidney injury and nephropathy. [13][14][15][16] The possible mechanisms by which Puerarin exerts its protective effects, at least in part, are related to its ability to increase superoxide dismutase activity, decrease the lipid peroxidation level and enhance fibrinolysis. [15][16][17][18][19] Furthermore, the underlying mechanisms by which Puerarin plays its renal protective role in the diabetic rats are unclear.…”

“…IKK specifically phosphorylates the protein that inhibits NFkappaβ, IKβα, resulting in its dissociation from NFkappaβ and, thereby allowing NF-kappaβ to migrate to the nucleus to generate the transcription of proinflammatory and adhesion molecules. Cocaine, GSK-3β, oxidative stress and high glucose may activate NFkappaβ to induce matrix metalloproteinase-9 (Kenny, 2011;Zhong and Kowluru, 2013;Zhong et al, 2014) which is pro-apoptotic and may increase sensitivity to pentylenetetrazole-induced seizures (Wilczynski et al, 2008). Metformin attenuates the cytokine expression of pro-inflammatory and adhesion molecule genes by inhibiting NF-kappaβ via AMPK activation (Hattori et al, 2006).…”

“…It may be crucial for epileptogenesis . In hyperinsulinaemia, oxidative stress, high glucose and GSK-3 beta may activate nuclear factor-kappaβ to induce matrix metalloproteinase-9 (Zhong and Kowluru, 2013;Zhong et al, 2014) which increases excitability to leptazol-induced seizures (Wilczynski et al, 2008). Nonspecific inhibitors of glycogen synthase kinase such as metformin and cannabinoids may suppress the production of interleukin-I beta, interleukin-6, interleukin-12 and tumor necrosis factor whilst enhancing production of interleukin-10.…”
